Study basis or key toxicological reasoning for E464

In 90-day studies, rats tolerated HPMC up to 9,000 mg/kg body weight per day and dogs up to 1,500 mg/kg per day. Dog groups contained only two animals of each sex per dose [1]. In a 91-day rat study, EFSA set the No Observed Adverse Effect Level (NOAEL) at 2,100 mg/kg per day because lower top-dose weight and food intake were not statistically significant [2]. One- and two-year rat studies tested up to 10,000 mg/kg per day. High-dose growth reduction and blood changes occurred, but no adverse tissue findings or tumour increase were identified [3]. Direct E464 genotoxicity and reproductive studies were unavailable, so EFSA used read-across from related celluloses [3]. At 5 g/day, mostly mild digestive complaints occurred [4], while trials using 5 or 15 g/day reported minimal adverse effects [5]. EFSA retained the propylene chlorohydrin limit because genotoxic risk could not be excluded [6]. One 2026 case in a patient with chronic urticaria and other hypersensitivities was supported by skin tests and an 8 g challenge. This single high-dose case does not represent normal food exposure [7].

  • No expected effect at normal food levels. E464 is minimally absorbed and has low oral toxicity.
  • Digestive symptoms at gram-level intake. A 5 g/day study reported mostly mild gas, bloating, loose stools and increased bowel movements [4].
  • Rare immediate hypersensitivity. One oral case linked HPMC to urticaria and angioedema after positive skin and challenge tests [7].
  • Manufacturing impurity control. EU specifications limit total propylene chlorohydrins to 0.1 mg/kg [6].

Most consumers do not need to avoid E464 in ordinary foods. Gram-level fibre products should be introduced gradually with enough fluid, and persistent gastrointestinal discomfort warrants reducing or stopping intake. Anyone who develops hives, facial swelling or repeated reactions after a hypromellose capsule, supplement or drink should stop the suspected product and seek medical assessment. The impurity concern relates to manufacturing quality and does not mean compliant E464 is genotoxic.

What is E464 – Hydroxypropyl methyl cellulose food additive?

E464 is a synthetic cellulose ether made from purified cellulose obtained from fibrous plant material, commonly wood pulp or cotton linters. Manufacturers treat the cellulose with alkali and react it with methyl chloride and propylene oxide so that some hydroxyl groups become methyl and hydroxypropyl groups. This modification lets the material swell and disperse in water, producing a viscous colloid. Different substitution patterns determine how each commercial grade behaves.

The cellulose backbone is not substantially digested or absorbed, so E464 acts mainly as a viscous, non-digestible polymer. Molecular size and substitution can be adjusted to create grades with different viscosity, gel temperature and film-forming properties. It is also non-ionic and remains useful across a broad range of food acidity.

In food, E464 binds water, thickens mixtures, stabilizes emulsions and suspensions, limits ice-crystal growth and forms films. It can gel when heated, which helps gluten-free dough and plant-based foods. The name hypromellose is common on pharmaceutical and supplement labels, especially vegetarian capsule shells.

Where is E464 – Hydroxypropyl methyl cellulose food additive allowed (EU vs US)?

The European Union authorises E464 in many categories and permits up to 10 mg/kg for marking citrus fruit, melons and pomegranates. EU specifications limit propylene chlorohydrins and each of arsenic, lead, mercury and cadmium to 0.1 mg/kg. In the United States, 21 CFR 172.874 permits HPMC as an emulsifier, film former, protective colloid, stabilizer, suspending agent or thickener under good manufacturing practice.
